What Is P-21? The CNTF-Derived Peptidomimetic in Neurogenesis Research

What Is P-21? The CNTF-Derived Peptidomimetic in Neurogenesis Research

P-21, also written P021 in the primary literature, is a small synthetic peptidomimetic designed to reproduce part of the biological activity of ciliary neurotrophic factor (CNTF), a cytokine in the same structural family as interleukin-6 that signals through the CNTF receptor complex. CNTF itself is a full-length protein that is difficult to deliver across the blood-brain barrier and has a short circulating half-life, which is the practical problem P021 was designed to work around: it is a much smaller molecule intended to engage downstream CNTF-associated signaling while being easier to produce and administer to research animals by mouth or by injection. The compound has been studied largely by one research group, based at the New York State Institute for Basic Research, across more than a decade of publications in rodent models of aging, Alzheimer's disease pathology and a genetic neurodevelopmental disorder model.

What is CNTF and why design a mimetic of it?

Ciliary neurotrophic factor is a cytokine that signals through a receptor complex shared with other members of the IL-6 family, activating JAK/STAT and related intracellular signaling cascades in neurons and glia. In rodent models, CNTF itself has been associated with neuroprotective and neurogenic effects, but as a full-length protein it does not cross the blood-brain barrier efficiently and is rapidly cleared from circulation, limiting its usefulness as a research tool for chronic dosing studies. P021 was developed specifically to reproduce a portion of CNTF's downstream signaling with a molecule small enough to be given systemically, including by the oral route in some published protocols, which is a major reason it has been studied as a distinct research compound rather than as a stand-in for CNTF protein itself (Kazim and Iqbal, 2016).

What have cell and early rodent studies examined?

The earliest published characterization of this class of compound examined neurotrophic peptides incorporating an adamantane group, reporting that these molecules improved performance on learning and memory tasks in mice and were associated with markers of neurogenesis and synaptic plasticity in hippocampal tissue (Li et al., 2010). Later work from the same research program used cortical and hippocampal neuron cultures alongside in vivo rodent testing to characterize P021 specifically as a CNTF-pathway small-molecule mimetic, reporting effects on neurite outgrowth and synaptic protein markers in cultured neurons that paralleled effects seen in treated animals (Kazim and Iqbal, 2016).

What has research examined in models of Alzheimer's disease pathology?

Several studies have examined P021 in the triple-transgenic mouse model of Alzheimer's disease, which develops both amyloid-beta plaques and tau pathology with age.

Cognitive-aging and hippocampal markers

Bolognin and colleagues examined chronic administration of P021 in aged, cognitively impaired rats, reporting improved performance on hippocampus-dependent memory tasks alongside increased markers of neurogenesis in the dentate gyrus and changes in synaptic protein expression relative to untreated aged controls (Bolognin et al., 2014).

Amyloid-beta and tau pathology

Baazaoui and colleagues reported that early, extended administration of P021 in triple-transgenic Alzheimer's disease model mice was associated with lower amyloid-beta and tau pathology burden, along with reduced neurodegeneration markers and better cognitive-task performance compared with untreated transgenic littermates (Baazaoui et al., 2017). A related study from the same group looked specifically at dendritic spine density and synaptic protein markers under the same treatment paradigm, reporting that P021 administration was associated with preservation of dendritic and synaptic structures that are otherwise lost as the pathology progresses in this model (Baazaoui et al., 2017).

Cerebrospinal fluid tau

Khatoon and colleagues measured tau protein levels in aged rat cerebrospinal fluid and reported that chronic P021 administration was associated with a reduction in elevated CSF tau relative to untreated aged animals, proposing CSF tau as a translatable biomarker readout for this line of research (Khatoon et al., 2015).

What has research examined outside Alzheimer's disease models?

A more recent study examined P021 in an in vitro and in vivo model of CDKL5 deficiency disorder, a genetic neurodevelopmental condition, reporting effects on neuronal morphology and behavioral measures in a mouse model carrying the relevant genetic mutation, extending this compound's study beyond aging and Alzheimer's disease models into a distinct neurodevelopmental research context (Mottolese et al., 2024). Taken together with the aging and Alzheimer's disease literature, this body of work treats P021 as a general tool for probing CNTF-pathway-dependent neurotrophic signaling across multiple rodent disease models rather than a compound tied to one specific pathology.

How does P-21 compare to other neurotrophic-pathway research peptides?

P-21 sits alongside other small peptides studied for effects on neuronal signaling and plasticity, though each engages a distinct receptor system; for background on an angiotensin IV-derived peptide studied in a separate cognitive-research context, see what is Dihexa. Researchers comparing across this category generally note that mechanism, receptor target and delivery route differ substantially even when the downstream readouts, such as hippocampal markers or cognitive-task performance in rodents, look superficially similar. Short regulatory peptides studied for tissue- and organ-specific signaling in a related but mechanistically distinct line of research are covered in bioregulator peptides explained.

Handling notes

P-21 is supplied as a lyophilized powder for research use. Like other short peptides, it should be stored per the certificate of analysis and reconstituted using standard bench technique and concentration math; this is bench math for preparing a solution for an assay, not guidance for use in any subject. See peptide half-life explained for background on why cold storage and prompt use after reconstitution matter for a given research timeline. As with any research peptide, confirm identity and purity from the lot-specific certificate of analysis before use in an assay. Lot-specific specs for research-grade P-21 are posted on our P-21 lot page.

Frequently asked questions

Is P-21 the same molecule as CNTF?

No. P-21, also written P021, is a synthetic small-molecule peptidomimetic designed to engage CNTF-associated receptor signaling. It is not the CNTF protein itself and is structurally much smaller than the full-length cytokine.

What animal models have been used to study P021?

Published studies have used aged rats, triple-transgenic Alzheimer's disease model mice, and a CDKL5-deficiency disorder mouse model, alongside cortical and hippocampal neuron cultures.

What outcome measures do these studies report?

Reported measures include hippocampal neurogenesis markers, dendritic spine and synaptic protein density, cerebrospinal fluid tau levels, amyloid-beta and tau pathology burden, and performance on hippocampus-dependent memory tasks in rodents.

Has P021 been studied in humans?

The published literature on P021 consists of cell culture and rodent studies. This article summarizes laboratory research models, not clinical evidence, and nothing here describes a protocol for use in any subject.

Why is P021 studied instead of CNTF protein directly?

CNTF is a full-length cytokine with a short circulating half-life and limited ability to cross the blood-brain barrier, which restricts its usefulness as a chronic-dosing research tool. P021 was designed as a smaller, more stable molecule intended to engage related signaling pathways for laboratory study.

How is P-21 supplied for laboratory use?

As a lyophilized powder intended for research use only, sold by lot with an accompanying certificate of analysis.
